If I create a custom D play -- say a play to try to contain a team's speedy RB from running to the outside -- and in the play AI it lets me tag a certain kind of RB, do I need to do anything else specific to that opponent? Or will it automatically say "if that type of player is in the game and the other inputs match" I'll run this defensive play?

You have to go to the team and then click "scout team" then tag the RB as speedy.

You have to tag the actual player and you do that by going to the opponents team page and then click on "scout team". You then select "your team" in the drop down menu (you have to have scout access from the owner to do this).
Once you select your team in the drop down menu then you click the "tag" link next to the player you wish to tag and select speedy or power or whatever matches your AI input.

You also have to ensure you don't have a play higher in your DAI that still fire during that look.
For example:
You built an input called "Strong I" and in that input you left all of the drop downs alone except the one where you selected the formation as "strong I". Below it you have an output that calls a generic play for Strong I formation...I would title an input like this "Strong I Default"btw.
If below it you create an input called "Strong I Speedy HB" and in the input you selected "Strong I" in the formation dropdown, and "Speedy" in the HB dropdown with an output that helped defend outside running, your tagged HB input would never fire; the default above it would because that input was all inclusive (for strong I plays).
Always put your default type inputs at the bottom of your list for each formation. The more specific you get, those have to be higher. So off you also added an input with Strong I selected, speedy HB selected AND blocking TE selected, that would have to be placed above both the Strong I Default and Strong I speedy HB.
Hopefully that makes sense.

lol probably true. It might make more sense if you were to open an AI and follow along as you read it. Probably should have just saved 10 minutes and did screen shots.
