"Slot Machine" says that the bonuses are applied when "running a rout". Does that mean that I have to be set to "Run exact rout" and If I am "Creative" I wont get the bonus?

you are still running a route when on creative... in fact you will run the exact same route as you would if you were on exact... you just run it a bit differently. so you will get the bonus regardless of your route running tactics.
