How much stamina do I need to play a full game effectively at level 16? Is 20 enough?

If you split time equally, 20 is probably enough.

If you are going to try and get almost all the plays on offense I would suggest getting stamina up to around 30 over the course of the season. However game 1 will tell you a LOT about what you need as the new energy system means the energy you have after a game will now be exactly how much your player had when the game ended. Decide how low you are comfortable letting your QB energy drop during a game and then keep increasing stamina until he consistently finishes above that number after games.

in my experience upper 30's. But that is also set by energy settings by the oc.
i have seen a 38 stam qb take all but 3 snaps in a game, fun times cause my guy was the backup. lol.

My QB played every single snap last season on hard intensity and finished practically every game at 80+ energy (I can't remember seeing 79 or lower, but can't say for sure he didn't do it at all) while averaging 73.1 plays/game. He ended the season at level 28 with 37 stamina without me doing much to increase it over the course of the season so I doubt he gained more than a point MAYBE 2 in that time.

Yeah, you want to eventually get your stamina to the upper 30s and even past 40, but at his level, 30 should be fine.
