If you are in a league, you get 16 league fights and 16 regular fights in a day, and you get $ for both. So getting in a league asap is important to get you extra $.

Once you are in a league, you go to any other league and look at the roster. When in the league roster page, if you click the boxing gloves, you are doing an interleague challenge.

It says the following under the preview for the two fighters
"This is a League battle between GLB - US Pro and GLB Network Fighters."

You can also fight players in your own league as well. You get 16 of these two types of fights combined. The first raises your league ranking. The second raises your own personal ranking inside your own league.
