Using your own experiences, what position is best used for STOP's? O-Line because you can mold them how you want and they are cheaper to make, or a defensive position since you need to make sure the opposing team first score on kick returns more than you need to make sure you do score on kick returns?

I no longer believe in STOPs, so I would want a FB that plays BW, and a LB built to create fumbles. Both players are therefore also useful outside of ST.

All STOP dots have the same major and minor attributes, no matter the position. The only areas where position makes a difference are with your available Special Abilities, height, and weight.
That's why it's so popular to just use O-line positions for STOP-dots... nice and cheap that way.
A 200-flex defensive dot for a STOP-dot is more expensive, but you'll get SAs that help you a little more at moving and tackling, plus you'll be lighter and thus a little faster.

I figured that it mostly depended on weight for speed, which is why I see some Safety's and Linebacker's instead of all O-Line.

STOP centers, IMO. Reason isn't about cost (but it's a bonus)... nor about the ability to mold as needed (can do that with any STOP archetype)... or about scoring/stopping scores on returns. It's about what you have to work with as a team.
Lets' say you have NO STOP archetypes on your team. What do you use? For stopping returns you have plenty of defensive players to choose from without hurting your "team defensive stamina"... and you can go double and even triple deep on the depth charts. But, when returning the ball, you have... of quality... with luck... 1 or 2 FB's you can use that will really work, 1 or 2 TE's you can use... and... um... some really slow O-linemen (mostly... the pass blocking archetypes do better than the run blocking just because they generally have more speed)... or some WR's that generally have as much strength as they were born with (around 8) and about the same in blocking. IF... if you're lucky, maybe your team likes the Power Receiver archetype and you can use them.
What I'm saying is, it is MUCH harder to fill the offensive side of the ST lineups than it is the defensive side of ST's. STOP centers (I like to have 4 myself) fill that void nicely, and... if built right, not only do great for your returns, but also on stopping your opponents returns.

personally I actually split my stops up. I will build 2 DE's for the gunner's. they are built similar to a pass rusher with a little more strength to help with FF's. the rest of the ST's unit are TE's most of the time. yeah they cost a little more but they make dang good blockers for the return game. I have used FBs and O-linemen also but I like the TEs and Fbs a little better.
Curious why you would use a 5 major position (TE) over a 4 (FB) for your blocker
