How should I make them able to get sacks or help defend the run

In a more traditional approach high strength, along with fairly high agility and a fair amount of speed would make you good against the run, and high agility and/or speed along with a fair amount of strength and maybe some jumping would make you good against the pass. But in the GLB world you could have the best run build in GLB and running backs would run right over you. Or you could have the best passing rush build and face a below average opponent and still not be able to create pressure on the QB.

Ummm, no. In a 3-4, the d-lines primary responsibility is to occupy blockers to free up LBs to make plays.
